Charley Hoffman withdrew from The American Express during Round 3 with a back injury, the Tour announced.
The 44-year-old was only three shots off the lead at 8-under par going into Saturday's rounds after shooting 70-66 in Days 1 and 2.
This move marks his first withdraw since the 2018 AT&T Pebble Beach Pro-Am.
Hoffman finished inside the top 15 at last week's Sony Open in his first start of 2021.
